Cryo-electron microscopy for integrative structural biochemistry

The course covers all typical steps of Single Particle Cryogenic Electron Microscopy (Cryo-M), from specimen preparation to atomic model building. Requirements of the sample for cryo-EM, major and most up-to-date software for performing single particle cryo-EM, as well as how Cryo-EM compares with and can be combined with other structural biology techniques are included. The course will also introduce other applications of cryogenic electron microscopy, such as cryo-electron tomography and electron diffraction of nano protein/compound crystals (microED).

  • Course structure

    After completing the course, the student is expected to be able to:

    - Explain the theory behind Cryo-EM

    - design a single particle cryo-EM project and identify possible pitfalls in the process

    - perform a Cryo-EM experiment, from specimen preparation to model building of a target protein

    - Show ability to analyse dynamic systems and use a combination of prediction methods, including AlphaFold, and manual model building.

    Modules

    The course consist of the following modules;

    Module 1: Teori (theory), 5 hp

    Module 2: Laboratory exercises (Laborationer), 2.5 hp

    Teaching format

    Teaching consist of lectures, group instruction, seminars, exercises and laboratory sessions.

    Assessment

    The course is examined in the following manner:

    Assessment of module 1 takes place through written exams, written reports and oral presentations.

    Assessment of module 2 takes place through written reports and oral presentations.
